Staff Scientist

The National Human Genome Research Institute, a major research component of the National Institutes of Health (NIH), and the Department of Health and Human Services (DHHS), is seeking exceptional candidates for a Staff Scientist position in the Organic Acid Research Section (OARS), Metabolic Medicine Branch (MMB).

About the position

OARS research focuses on the genetics, molecular mechanisms, pathophysiology and treatment of the hereditary disorders of propionylcoa oxidation, including isolated methylmalonic acidemia (MMA), propionic acidemia (PA), and disorders of intracellular cobalamin metabolism. Candidates must have direct experience working with multiple forms of acylation modifications especially methylmalonylation, malonylation, succinylation, glutarylation, acetylation, and propionylation and their effects on target protein function. Applicants must also have experience in cloning, testing, and optimizing AAV vectors, working with mouse models of human disease, and working with primary and immortalized cell lines. Primary responsibilities will include but are not limited to in vitro enzymatic assays such as deacylation assays with class III histone deacylases; in vitro non-enzymatic and enzymatic acylation assays; gene ontology analysis and validation of large tandem mass spectrometry data sets; immunoblotting and protein immunoprecipitation/co-immunoprecipitation assays on cell lines, murine tissues and human clinical samples; AAV vector cloning and design; AAV gene therapy administration to cell cultures and murine models; creation and characterization of genetically engineered mouse models; mentoring junior staff and demonstrated ability to collaborate. Experience with the preparation of patent applicants is desirable.

Qualifications

Candidates must have a Ph.D. and minimum of five years’ experience in genetics, metabolism, and molecular biology and related experiments with specific expertise in post-translational modifications, mouse modeling, biochemistry, molecular biology, and AAV gene therapy.
